Promoter is the site on the DNA to which an RNA polymerase molecule binds prior to initiating transcription. The promoter also contains the information that determines which of the two DNA strands will be transcribed, that is, it defines template and coding strands. Based on the position of promoter, template and coding strands are defined. Promoter is located upstream of structural gene towards its 5′ end (with reference to the coding strand). Terminator is hence located downstream of the coding strand towards it 3′-end (with reference to coding strand)
